私たちの研究室では、このような生命分子をハンドリングする技術的な充実を背景として、臨床診断、食品・環境計測をはじめとする分析分野への応用、物質・エネルギー生産、さらには低炭素社会の実現を目指して、生命分子のエンジニアリングを通した合目的な生命分子の創出とこれを応用した新産業技術としてのバイオデバイス、バイオプロセスの開発を進めています。

1. 生命分子工学
多様な蛋白質設計・シュミレーションツールを駆使し、高機能で合目的な次世代酵素・蛋白質分子の開発を進めています。例えば、バイオツール素材として適切な耐久性が求められる蛋白質の安定化、臨床診断での夾雑物の影響を排除するための基質特異性の改変はもとより、直接電子移動型酵素の創製、結合蛋白質と酵素を融合した新規分析素子、酸化酵素から脱水素酵素への改変といった新しい酵素改良の戦略も提言しています。

2.バイオセンシング技術開発
生命分子の優れた分子認識能力を活用した計測技術であるバイオセンシング技術は、糖尿病患者における自己血糖測定(Self Monitoring of Blood Glucose; SMBG)装置に代表され、医療診断技術としての地位を確立しています。私たちの研究室で開発された分子認識素子としての優れた能力を有する生命分子を活用することで、独創的で産業競争力に富む新しい計測技術や計測装置の開発を目指しています。例えば、直接電子移動能力を有する酸化還元酵素と微小電極を用い、皮下埋め込み型の連続グルコース計測装置の開発も進めています。例えば、外部電源を必要とせず血液中のグルコースだけで血糖値を計測、データを送信するワイヤレスグルコースセンシングシステムであるBiocapacitorや自立型のセンシングアクチュエータの開発を進めています。すなわち、薬剤ポンプを駆動するエネルギー源として血液中のグルコースを用い、その濃度により駆動を制御し、グルコース濃度に応じて例えばインスリンを投与するバイオデバイスの開発を進めています。

3.合成生物学のコンセプトに基づく新規バイオプロセス開発
生命科学に関する支援技術はめざましい進歩を遂げており、合成生物学の考え方を取り入れた新しいバイオプロセス設計が現実味を帯びてきています。私たちは、光合成能力を有する原核生物、藍藻(シアノバクテリア)の有する優れたバイオ燃料関連化合物生産能力に注目した「シアノファクトリ」(JST戦略的創 造推進事業(CREST)研究代表)の開発を推進しています。シアノファクトリは合成生物学のコンセプトに基づき、人工的にプロセス制御できる合成情報伝 達系が組み込まれた海洋合成シアノバクテリアホストを開発・応用することで、バイオ燃料関連化合物の生合成から目的化合物の回収プロセスまで一貫してデザ インされた、バイオ燃料関連化合物生産の全く新しいバイオプロセスの実現をめざしています。本研究プロジェクトは、東京農工大学工学府生命工学専攻の主要な研究者の協力のもと推進されている専攻横断的な研究テーマです。私たちの研究室は、本プロジェクトの代表研究室であるとともに、藍藻における新しい光セ ンシングインターフェイス分子を開発することで、光信号により藍藻の凝集・溶菌が容易に制御できるバイオエネルギー化合物生産バイオプロセスのために設計された合成海洋藍藻宿主を開発することを目的としています。
また、我々の研究室はInternational Genetic Engineering Machine Competition (iGEM)の本学チーム”Tokyo-NoKoGen”のホスト研究室として、同チームの活動をサポートしています。

Summary

Our research group is engaged in the development of innovative biosensing technologies dedicating for healthcare, and also novel bioprocesses based on synthetic biology approaches. Our strategic research concept is to create novel molecules based on the biomolecular engineering to be applied for the development of emergent industrial technologies.

1. Biomolecular engineering
We have been engaged in the development of variety of oxidoredctases, which have been utilized for electrochemical biosensors. Recently, we are focusing on the creation of dehydrogenases by engineering oxidases, which is now possible by detailed investigation of the oxidative half reaction of these enzymes. Our interests are not limited to oxidoreductases, but also several kinases, binding proteins, and antibodies are involved. Protein structural analyses are also carried out toward industrially significant enzymes, such as fungus derived FAD glucose dehydrogenase which is the current most popular and advanced enzyme to be utilized in the blood glucose monitoring.

2. Biosensing technology
Varieties of biosensing technologies are currently studied and developed in our group, combined with our original biomolecules, mainly based on electrochemical principles; from conventional amperometric enzyme sensors to direct electron transfer principle based autonomous self-powered sensing devices, designated as BioCapacitors. As the plat-form technologies, we use our original disposable electrodes (carbon, gold) with variety of features, from conventional screen printed electrodes to interdigitated electrode array. Our goal is to develop sensors dedicating for personal and professional healthcare management.

3. Synthetic biology
Our research interest is to develop plat-form technologies for bioprocesses, based on synthetic biology approaches. We are currently engaged in the project based research, designated as “CyanofactoryTM” (Sode serves as the PI). The CyanofactoryTM is composed of 1) synthetic marine cyanobacterial host strains, 2) synthetic operons for the production of biofuel-related compounds, and 3) the employment of ionic liquids for downstream processing. Our group is engaged in the development of synthetic marine cyanobacterial host strains, throughout the development of artificial signal transduction system by designing the light sensing gene expression system using two component regulation system and riboregulators. The technologies are dedicated not only for the designing of bioenergy production processes, but also for heterotrophic bacteria based bioprocesses for chemical.
